The Thyroid Transcription Factor 1 antibody identifies the 40 kDa TTF-1 band in nuclear extracts or whole-cell lysates of TTF-1-positive cell lines. It is a member of the NKX2 family of transcription factors expressed in epithelial cells of the thyroid and lung. Nuclei from liver, stomach, pancreas, small intestine, colon, kidney, breast, skin, testis, pituitary gland, prostate, and adrenal glands are non-reactive. TTF-1 is detected in primary adenocarcinomas and small cell carcinomas of the lung and is absent in colon and breast carcinomas. Staining with the TTF-1 antibody is useful for distinguishing between lung tumors and tumors of non-lung origin. In addition, it can also be used to differentiate small cell lung carcinomas (SCLC) from lymphoid infiltrates. Loss of TTF-1 expression in non-small cell lung cancer (NSCLC) has been associated with the aggressive behavior of such neoplasms.
