There are four human SALL proteins (SALL1, 2, 3, and 4) with structural homology that play diverse roles in embryonic development, kidney function, and cancer development. SALL4, also known as Sal-like protein 4, is a zinc-finger transcription factor for the maintenance of pluripotency. Together with other pluripotency-related transcription factors such as OCT4 and NANOG, it is a master regulator of embryonic pluripotency. SALL4 expression is low to undetectable in most adult tissues, with the exception of germ cells and human blood progenitor cells. However, it is reactivated and dysregulated in various cancers, such as acute myeloid leukemia, acute B-cell lymphocytic leukemia, gastric cancer, breast cancer, hepatocellular carcinoma, lung cancer, and glioma. Expression of SALL4 in germ cells makes it a useful marker for germ cell tumors such as seminomas, embryonal carcinomas, yolk sac tumors, and teratomas. SALL4 expression is often associated with poorer survival and prognosis, such as in hepatocellular carcinoma, or with metastasis, such as in endometrial carcinoma, colorectal carcinoma, and esophageal squamous cell carcinoma.
