NKX2.2 is a transcription factor that plays a critical role in controlling cell fate specification and differentiation in many tissues. In the developing central nervous system, this developmentally important transcription factor acts as a transcriptional repressor, controlling oligodendrocyte differentiation and myelin gene expression. The role of the various NKX2.2 structural domains in this process is unclear. NKX2.2 expression has been found in intestinal neuroendocrine tumors, making it a potential marker for the study of gastrointestinal neuroendocrine tumors. Recently, the NKX2.2 protein was identified as a target of EWS-FLI-1, the Ewing sarcoma-specific fusion protein, and array-based gene expression analyses have shown that it is differentially upregulated in Ewing sarcomas. It acts as a valuable marker for Ewing sarcomas and helps in the differential diagnosis of small round cell tumors.
