CD3 is a protein complex consisting of five chains (gamma, delta, epsilon, zeta, eta) associated with the T-cell receptor (TCR). CD3e (T-cell surface glycoprotein CD3 epsilon chain) is a membrane protein that plays a role in the assembly of the TCR-CD3 complex and in signal transduction, which regulates T-cell proliferation, apoptosis, and thymocyte development. Some of the signal-transducing molecules recruited by CD3e are oncogenic, leading to disruption of CD3e signaling. Overexpression of CD3e is associated with a high incidence of T-cell lymphomas, whereas low or absent CD3e expression has not been associated with lymphomas. Defects in CD3e are associated with immunodeficiency. QR004 may be helpful in the detection of T cells and T cell-derived malignancies such as multiple myeloma, acute myeloid leukemia, or chronic lymphocytic leukemia. CD3e in combination with CD56 is a characterization marker that distinguishes NK cells from other immune cells. Phenotypic expression of CD3e and CD56 by NK cells is also a very useful marker for extranodal NK/T cell lymphoma (ENKTCL), a highly aggressive mature NK/T cell neoplasm.
