B lymphocytes produce immunoglobulins consisting of two identical heavy chains and either two identical kappa light chains or lambda light chains. Normal lymphatic tissue therefore contains a mixture of B cells expressing kappa and lambda light chains in a 2:1 ratio. This ratio is no longer present in B-cell tumors, as they arise from a single transformed cell and therefore express only one type of light chain. The monoclonality of malignant lymphomas is used as a diagnostic marker when detected with lambda light chain antibodies. This antibody labels the lambda light chains of human immunoglobulins expressed by B lymphocytes and plasma cells. It can be helpful in the detection of leukemia, plasmacytomas, and various non-Hodgkin lymphomas. Other cells may also express lambda light chains, which arise from nonspecific immunoglobulin uptake. The lambda light chain antibody can be used in a panel with the kappa light chain.
