This pseudotyped virus uses lentiviral vectors to package Spike (S) protein of SARS-CoV (Severe Acute Respiratory Syndrome, GenBank: DQ412594.1), which was designed by codon optimizing the full-length S sequence for mammalian expression. Infection of cells with this pseudotyped virus carrying luciferase reporter results in detectable luciferase activity. See our titration result showing the undiluted pseudovirus (green) generates signal 10,000-fold higher than uninfected control (cell alone as background in blue). We also evaluated the neutralizing activity of SARS Positive control mAb and 2 published neutralizing mAbs (CR3022 and 80R) by using this pseudotyped virus, see results.
